Der Pathologe Koss konzentrierte sich in seinen wissenschaftlichen Arbeiten auf die Krebsforschung an verschiedenen Organen. Hierbei legte er den Schwerpunkt auf die Geschwulstentwicklung im Gebärmutterhals, in der Gebärmutterschleimhaut sowie der Harnblase. Als Pionier auf dem Gebiet der Zytodiagnostik verband er darüber hinaus histologische mit zytologischen Ansätzen zur Erweiterung von Diagnoseverfahren.

Leopold G. Koss dedicated his professional endeavors to the study of human cancer, its diagnosis and natural history. He was a pioneer in the field of cytologic diagnosis of cancer of various organs with special emphasis on the uterine cervix, endometrium and the urinary bladder. In 1956 he described in detail cell changes caused by human papillomaviruses in the uterine cervix (koilocytosis). He also determined the important role of silent carcinoma in situ in the natural history of bladder cancer. He was a pioneer in image analysis of cells and, within the recent years, studied the arrangement of chromosomes in normal human nuclei. He is the author of "Diagnostic Cytology and its Histopathologic Bases", 12 other books and 395 scientific papers.
